Isothermic non PCR procedure

Definition: a primer dependent technology that can be used for the continuous amplification of nucleic acids in a single mixture at one temperature

Isothermal nucleic acid amplification of RNA is achieved by the simultaneous action of avian myeloblastosis virus reverse transcriptase (AMVRT), T7 RNA polymerase and RNase H

Nucleic acid sequence based amplification (NASBA) was developed in the early 90s to amplify nucleic acids without the help a thermal cycler. This technique mainly used to obtain many copies of RNA starting from a few RNA molecules

• avian - طيور - عثمان كان حمامة او طير المسجد
• تم تطوير هذه التنقية سنة 1990 لمضاعفة الـ RNA بدون استعمال جهاز Thermal cycler 
• فائدته هي تضخيم عدد الـ RNA بدئا بعدد قليل من جزيئات الـ RNA

• Immediately after the invention of NASBA, it was used for the rapid diagnosis and quantification of HIV-1 in patients` sera
• Quicker than PCR
• Isothermal
• More sensitive
• Used to detect pathogenic viruses with ssRNA genomes Eg: influenza A, Foot and mouth disease virus, SARS, HboV, Trypanosoma brucei
